A.P.Polo - Incubator 2 - Hamburg / Germany. An incubator is a device used to grow and maintain microbiological cultures or cell cultures. The incubator maintains optimal temperature, humidity and other conditions such as the CO2 and oxygen content of the atmosphere inside. Incubators are essential for much experimental work in cell biology, microbiology and molecular biology and are used to culture both bacterial and eukaryotic cells. An incubator is made up of a chamber with a regulated temperature. Some incubators also regulate humidity, gas composition, or ventilation within that chamber. The simplest incubators are insulated boxes with an adjustable heater, typically going up to 60 to 65 °C (140 to 150 °F), though some can go slightly higher (generally to no more than 100 °C). The most commonly used temperature both for bacteria such as the frequently used E. coli as well as for mammalian cells is approximately 37 °C (99 °F), as these organisms grow well under such conditions. For other organisms used in biological experiments, such as the budding yeast Saccharomyces cerevisiae, a growth temperature of 30 °C (86 °F) is optimal. More elaborate incubators can also include the ability to lower the temperature (via refrigeration), or the ability to control humidity or CO2 levels. This is important in the cultivation of mammalian cells, where the relative humidity is typically >80% to prevent evaporation and a slightly acidic pH is achieved by maintaining a CO2 level of 5%. The Dutch physicist, chemist, designer and instrument maker Cornelis Drebbel (1572-1633) is regarded as the inventor of the first thermostat of modern times, which forms the basis of the incubator. He developed it for alchemical ovens and incubators for hatching chicken eggs. In this incubator, a fire burned under the actual incubator, the hollow walls of which were filled with water. The combustion gases rose up the outside walls of the incubator and could escape through a flue. To regulate the water temperature, a glass temperature sensor was inserted into the water-filled base of the incubator. One cylindrical part of this elongated glass body was filled with alcohol, while a second, U-shaped part was filled with mercury. The mercury column moved with the water temperature due to the thermal expansion of the alcohol. With the mercury level on, a floating rod lying on top was moved. A lever connected to this closed or opened the smoke outlet and thus the oxygen supply, which was used to regulate the fire in the oven and thus also the temperature. In the 18th century, it was the French inventor and chicken breeder Jean Simon Bonnemain (~1743-1828) who was the first to invent an industrially producible temperature control - again for an incubator used in chicken breeding. It was based on the principle that different metals expand differently under the influence of temperature. An iron rod in a lead or zinc pipe acted as a temperature sensor in the water. The lead pipe is sealed at the lower end and the iron rod is firmly screwed to this seal. A copper or brass socket is soldered onto the upper end of the pipe and connected to the fresh air flap via a lever arm. As the coefficient of linear expansion of lead is greater than that of iron, the pipe expands more than the iron rod as the temperature rises, the fresh air damper closes and the temperature drops due to the falling oxygen supply. In the second half of the 19th century, Robert Koch and his institute staff in particular devoted themselves to the development of microbiological methods and techniques. In 1881, for example, he had an incubator built for cultivating bacteria. A short time later, incubators were already being mass-produced, most notably by the Berlin company Lautenschläger. Lautenschläger's "thermoregulator" (similar to Drebbel's design) was based on the thermal expansion of mercury, which was used in a closed system to regulate a gas burner. At this time, incubators were only operated at a constant temperature; it was not yet possible to regulate the temperature to several different levels. They were insulated with asbestos or linoleum, a water jacket ensured heat transfer to the interior and were heated with gas. Double doors, the inner ones made of glass, were also standard. However, calibrating the temperature regulator was much more complex than it is today. With the increasing electrification of cities, electrically heated incubators moved into laboratories from the beginning of the 20th century. However, the control principles remained mechanical for a long time and mercury was frequently used, as its thermal expansion is largely directly proportional to the temperature. With the invention of contact thermometers, these were the most common switching sensors in temperature control units until well into the 20th century. Initially, they had fixed contacts and could only switch an electric heater on or off via a relay. In 1926, the Ilmenau-based company Juchheim applied for a patent for the first glass contact thermometer, which enabled variable temperature settings via adjustable metal filaments. From the 1970s onwards, fully electronic controllers and later, with the advent of the digital age, microprocessor-controlled controllers in combination with highly sensitive temperature sensors ensured that temperature control units today have precise control behaviour. Resistance thermometers with platinum temperature sensors are usually used. Today's appliances enable a wide range of temperature curve control and temperature monitoring functions. They communicate with other devices or external computers and exchange data with device-specific or higher-level programmes. Many devices have internal data loggers in which all measured values and parameters are recorded in a tamper-proof manner over a certain period of time.
